What is Everolimus 10 mg?

Everolimus, sold under brand names like Afinitor, Afinitor Disperz, and Zortress, is a medication that belongs to a group of drugs known as kinase inhibitors. It’s used for two main purposes: preventing organ transplant rejection and treating certain noncancerous and cancerous tumors.

The specific brand and dosage of everolimus vary depending on the condition being treated:

  • Afinitor (available in 2.5 mg, 5 mg, 7.5 mg, and 10 mg tablets) and Afinitor Disperz (available in 2 mg, 3 mg, and 5 mg tablets for oral suspension) are typically used to treat certain types of tumors.
  • Zortress (available in 0.25 mg, 0.5 mg, 0.75 mg, and 1 mg tablets) is used to prevent the rejection of transplanted organs, like a liver or kidney.

Everolimus works by targeting a specific protein in the body called mTOR (mammalian target of rapamycin). This protein plays a key role in controlling how cells grow, multiply, form new blood vessels, and use nutrients.

When treating tumors, everolimus reduces the blood supply to the tumor, which helps slow its growth. It does this by binding to a protein called FKBP-12, forming a complex that blocks mTOR’s activity.

  1. Breast Cancer: It’s used for postmenopausal women with advanced hormone receptor-positive, HER2-negative breast cancer. It is given along with exemestane when other treatments haven’t worked.
  2. Pancreatic Cancer: Pancreatic is prescribed for adults with a type of pancreatic cancer known as pancreatic neuroendocrine tumor (PNET) that has progressed and can’t be treated with surgery.
  3. Neuroendocrine Tumors (NET): It helps adults with neuroendocrine tumors of the stomach, intestines, or lungs that have advanced and can’t be removed surgically. However, it’s not for those with carcinoid tumors that produce hormones.
  4. Kidney Cancer (RCC): RCC is for adults with advanced kidney cancer when other treatments have failed.
  5. Tuberous Sclerosis Complex (TSC)-related Tumors:
    • Kidney Tumors: It treats adults with a kidney tumor called angiomyolipoma, which doesn’t require immediate surgery.
    • Brain Tumors (SEGA): It’s used for adults and children (1 year and older) with subependymal giant cell astrocytoma (SEGA) that can’t be fully removed by surgery.

The Afinitor Disperz brand is specifically for:

  • Tuberous Sclerosis Complex (TSC):
    • Brain Tumors (SEGA): Treats adults and children (1 year and older) with SEGA when surgery isn’t an option.
    • Seizures (Epilepsy): Used as an additional treatment for adults and children (2 years and older) with certain types of seizures linked to TSC.

The Zortress brand of everolimus is used to prevent transplant rejection:

  • Kidney Transplants: It is given with other medicines like cyclosporine and corticosteroids to prevent rejection in people who’ve had a kidney transplant.
  • Liver Transplants: It’s also used with tacrolimus and corticosteroids for those who’ve had a liver transplant.

It’s important to note that the safety and effectiveness of everolimus have not been established in:

  • Children for treating breast cancer, neuroendocrine tumors, kidney cancer, or TSC-related kidney tumors.
  • Organ transplants other than kidney and liver.
  • Children under 18 years of age for the Zortress brand.

What should I do if I forget a dose?
